石内研究室 ── 極低温レーザー分光で見る分子認識の世界

理学院 化学系 石内研究室は,2020年5月1日に発足しました。

発足からしばらくの間,研究室は横浜(旧・すずかけ台)キャンパスにある科学技術創成研究院・化学生命科学研究所を拠点としていましたが,2023年5月に大岡山キャンパス本館へ移転しました。

ここでは,大岡山で本格的に始動した私たちの研究について,簡単にご紹介します。

The Ishiuchi Laboratory — Molecular Recognition Seen Through Cryogenic Laser Spectroscopy

The Ishiuchi Laboratory, in the Department of Chemistry, School of Science, was founded on May 1, 2020.

For some time after its founding, the lab was based at the Laboratory for Chemistry and Life Science, Institute of Innovative Research, on the Yokohama (formerly Suzukakedai) Campus, before relocating to the Main Building of the Ookayama Campus in May 2023.

Here we give a brief introduction to the research we have been carrying out in earnest since our move to Ookayama.

分子認識とは?

分子認識とは,生命を維持するために欠かせない機能のひとつです。 生体内には多種多様な分子が混在しており,その中で特異的な反応を引き起こすためには,無数の分子の中から特定の分子だけを選び取る機能分子認識が不可欠です。

例えば,遺伝情報はDNA塩基対どうしの分子認識によって正確に保存されています。 神経伝達物質やホルモンによる情報伝達も,それらと受容体タンパク質との間の分子認識によって成り立っています。 さらに,抗原・抗体反応のように自己と他者を区別するという,生物にとって最も基本的かつ重要な機能も,まさに分子認識そのものだといえます。

分子認識は,人工的に合成した分子でも実現できます。 その代表例がクラウンエーテルです。クラウンエーテルはアルキル鎖がエーテル結合でつながった環状分子で,金属イオンとキレート錯体を形成します。 このとき,環の大きさに応じて包接する金属イオンに選択性が生じることが知られています。

このイオン選択性は,一見すると単純な現象に思えますが,実際はそれほど単純ではありません。 周囲に溶媒分子が存在しない真空中では,このイオン選択性は失われてしまいます。 つまり,イオン選択性の発現には溶媒分子の関与が不可欠だということです。

このように,一見シンプルに見える分子システムの背後には,実は非常に複雑な仕組みが隠されています。 その仕組みを解き明かすためには,最先端の分光測定実験と計算化学によるアプローチの両方が必要となるのです。

What Is Molecular Recognition?

Molecular recognition is one of the functions essential to sustaining life. A vast variety of molecules coexist within living organisms, and triggering a specific reaction among them requires the ability to select just one particular molecule out of countless others — this is molecular recognition.

For example, genetic information is stored accurately through the molecular recognition between DNA base pairs. Signal transduction by neurotransmitters and hormones likewise relies on molecular recognition between these molecules and their receptor proteins. Furthermore, the antigen–antibody reaction — the most fundamental and important function by which organisms distinguish self from non-self — is itself a prime example of molecular recognition.

Molecular recognition can also be realized with artificially synthesized molecules. A representative example is the crown ether: a cyclic molecule made of alkyl chains linked by ether bonds, which forms chelate complexes with metal ions. Depending on the size of the ring, selectivity arises for which metal ion is encapsulated.

This ion selectivity may appear to be a simple phenomenon at first glance, but it is in fact far from simple. In a vacuum, with no surrounding solvent molecules, this ion selectivity disappears. In other words, the involvement of solvent molecules is essential for ion selectivity to emerge.

In this way, seemingly simple molecular systems conceal remarkably intricate mechanisms behind them. Unraveling these mechanisms requires both cutting-edge spectroscopic experiments and computational chemistry approaches.

冷却イオントラップ赤外分光 Cold Ion Trap Infrared Spectroscopy

冷却イオントラップ赤外分光とは,一言でいうと「孤立した極低温の分子の分光測定」です。 「孤立した」とは,周りに溶媒分子のない状態,すなわち宇宙空間に分子が1個だけ浮いているような状態を想像してください。

Cold ion trap infrared spectroscopy is, in short, "spectroscopic measurement of isolated, ultracold molecules." By "isolated," picture a state with no surrounding solvent molecules — as if a single molecule were floating alone in outer space.

このような状態では,溶媒分子が周りに存在しないときの分子そのものの性質を研究できるだけでなく,有限個の(個数を制御した)溶媒分子で溶媒和された状態を研究することもできます。 さらに極低温まで冷却することで,室温では様々な構造の間を揺らいでいる分子を「瞬間凍結」します。 これにより,室温状態では異性体として区別できない種々のコンフォメーションを異性体として区別できるようになり,柔らかな分子の構造化学研究が可能になります。

In such a state, we can study the intrinsic properties of a molecule with no solvent around it, and we can also study states solvated by a finite, precisely controlled number of solvent molecules. Cooling to extremely low temperatures further "flash-freezes" molecules that, at room temperature, fluctuate between many different structures. This lets us distinguish, as isomers, conformations that cannot be told apart as isomers at room temperature — enabling structural chemistry studies of floppy molecules.

このような状態を実現するのが冷却イオントラップ(図2 黄色い部品)です。 これは,真空中に導入した分子イオンを空間的に捕捉し,極低温に冷却する装置です。 イオントラップ自体は4Kまで冷却されていますが,トラップされた分子イオンはおよそ10K程度まで冷却されます。

The device that realizes this state is the cold ion trap (the yellow component in Figure 2). It spatially confines molecular ions introduced into a vacuum and cools them to extremely low temperatures. The ion trap itself is cooled to 4 K, while the trapped molecular ions reach roughly 10 K.

イオントラップには目的の分子イオンを溜め込むことができますが,通常の分光測定で用いる溶液などと比べると濃度は遥かに低く(~1 pmol/L),入射光と出射光の強度差から吸収を測る通常の吸収分光法を用いることは困難です。 このようなときに威力を発揮するのが「アクション分光法」です。 アクション分光法とは,光の吸収を出射光の強弱で検出するのではなく,光を吸収した結果として起こる別の現象を検出することで,吸収スペクトルに相当するスペクトルを測定する手法です。

The ion trap can accumulate the target molecular ions, but their concentration is far lower (~1 pmol/L) than that of the solutions typically used in ordinary spectroscopy, making it difficult to use conventional absorption spectroscopy, which measures absorption from the intensity difference between incident and transmitted light. This is where "action spectroscopy" proves its worth: rather than detecting light absorption through changes in transmitted-light intensity, it detects a separate event that occurs as a result of absorbing light, measuring a spectrum equivalent to an absorption spectrum.

我々は,水素分子をイオンに付着させ,吸収の際に水素が脱離することを検出することで,吸収スペクトルを測定しています。(水素タグ法) また,溶媒分子が脱離することを検出することでも,溶媒和状態の吸収スペクトルを測定できます。

We attach a hydrogen molecule to the ion and measure the absorption spectrum by detecting the loss of that hydrogen upon absorption (the "hydrogen-tagging" method). We can also measure the absorption spectrum of solvated states by detecting the loss of solvent molecules instead.
